Proteostasis of host cell showed a highly effect on SARS-CoV-2 replication. Compounds modulating this pathway prevented SARS-CoV-2 replication in cells. NMS-873, a small molecule inhibitor of the AAA ATPase p97 which is a key component of proteostasis affecting protein degradation, membrane fusion, vesicular trafficking and disassembly of stress granules, was found to inhibit SARS-CoV-2 replication at low nanomolar concentrations. |
