Action against Disease Model Prolarix The prodrug CB1954 (5-(aziridin-1-yl)-2,4-dinitrobenzamide) is activated by Escherichia coli nitroreductase (NTR) to a potent DNA-crosslinking agent. Virus-mediated expression of NTR in t uMour cells sensitizes them to CB1954 in vitro and in vivo, providing the basis for a strategy of cancer gene therapy. The Effect of Target Knockout, Knockdown or Genetic Variations NQO1 and NQO2 single knockout mice exposed to DMBA and BP are known to develop low frequency of skin t uMors as compared with WT mice (Fig. 2C; refs. 13-16). A comparison of DMBA- and BP-induced skin t uMors and t uMor multiplicities in DKO mice with WT, NQO1?/?, and NQO2?/? mice is plotted in Fig. 2C and D. DMBA exposure led to almost similar incidence of skin t uMors in individual NQO1 and NQO2 knockout and DKO mice that were significantly higher than WT mice (Fig. 2C). However, BP treatment showed significantly higher incidence of t uMors in DKO mice as compared with not only WT but also individual NQO1 and NQO2 knockout mice (P < 0.0001; Fig. 1C). Intriguingly, highly significant differences between DKO and individual NQO1 and NQO2 knockout mice exposed to DMBA and BP were observed in t uMor multiplicity (Fig. 2D). DKO mice exposed to DMBA developed 10 to 18 t uMors per mouse as compared with 3 to 4 t uMors per NQO1?/? and NQO2?/? mouse. Similarly, the exposure to BP led to four or more t uMors in DKO mouse as compared with a single t uMor in NQO1?/? and NQO2?/? mice. [531262]
